Thesis Title:

Exploring circadian physiology to inform mechanisms of myopia aetiology and management  

Thesis abstract:

Myopia (short-sight) is an eye condition associated with an increased risk of developing sight threatening eye disease. The rising global prevalence of myopia has increased the urgency to understand the causes of myopia and to develop effective anti-myopia strategies. Recent evidence suggests circadian biology may play a role in both myopia development and the mechanism by which bright light and time outdoors protects against myopia. The overall aim of this thesis is to explore aspects of human circadian physiologyincluding melatonin, melanopsin function, and sleep, in addition to external inputs to the circadian system, such as light exposure and physical activity, to inform mechanisms of myopia aetiology and management. Ocular biometrics, actigraphy, light sensors, sleep quality and chronotype questionnaires, and biochemical analytics will be used to explore relationships between refractive error, melatonin, circadian phase, light exposure, physical activity and sleep in young adult subjects. Additionally, the effect of atropine on melanopsin-driven pupil responses will be investigated using chromatic pupillometry.
